Human Cytomegalovirus Genome Diversity in Longitudinally Collected Breast Milk Samples

چکیده

Reactivation and shedding of human cytomegalovirus (HCMV) in breast milk during lactation is highly frequent HCMV-seropositive mothers. This represents a key transmission route for postnatal HCMV infection can lead to severe disease preterm neonates. Little known about strain composition or longitudinal intrahost viral population dynamics from immunocompetent women. We performed HCMV-specific target enrichment high-throughput sequencing 38 samples obtained Germany between days 10 60 postpartum 15 mothers with DNA lactia, assembled consensus sequences de novo . The genotype distribution number strains present each sample were determined by quantifying genotype-specific sequence motifs 12 hypervariable genes, revealing wide range genotypes (82/109) these genes the cohort unique, longitudinally stable mother. up three distinct was detected 8/15 mothers, indicating that representative subset woman’s reservoir might be locally reactivated early lactation. As described previously, nucleotide diversity multiple much higher than single strains. Breast as main source mother-to-infant may serve repository thus play an essential role natural epidemiology HCMV.

Cytomegalovirus in human breast milk: risk to the premature infant.

Human cytomegalovirus (HCMV) can be transmitted through breast milk to neonates. Although healthy full-term infants rarely develop symptoms of CMV infection; premature or low-birth-weight infants can experience symptomatic infection that is occasionally severe. There is limited information on the long-term effects of postnatal CMV infection in premature infants, suggesting that these infants do...

Metabolomic Approaches to Explore Chemical Diversity of Human Breast-Milk, Formula Milk and Bovine Milk

Although many studies have been conducted on the components present in human breast milk (HM), research on the differences of chemical metabolites between HM, bovine milk (BM) and formula milk (FM) is limited. This study was to explore the chemical diversity of HM, BM and FM by metabolomic approaches. GC-TOFMS and UPLC-QTOFMS were applied to investigate the metabolic compositions in 30 HM sampl...

Bacterial Composition and Diversity in Breast Milk Samples from Mothers Living in Taiwan and Mainland China

Human breast milk is widely recognized as the best source of nutrients for healthy growth and development of infants; it contains a diverse microbiota. Here, we characterized the diversity of the microbiota in the breast milk of East Asian women and assessed whether delivery mode influenced the microbiota in the milk of healthy breast-feeding mothers. We profiled the microbiota in breast milk s...

Diversity of human parechoviruses isolated from stool samples collected from Thai children with acute gastroenteritis.

A total of 82 fecal specimens which were known to be negative for rotavirus, adenovirus, norovirus, sapovirus, and astrovirus and which were collected from infants and children with acute gastroenteritis in Chiang Mai, Thailand, from January to December 2005 were screened for human parechovirus (HPeV). HPeV was detected by reverse transcription-PCR with a primer pair that amplified the 5' untra...
